blocking some external addresses

Good afternoon,

Is there any way in Kopano (other than changing the MTP directly) to block clients from sending emails to a particular email address.

Or the only way is to do it on the MTP side?

I would do it exactly like that. The only effective block is to not accept these mails, and therefore rejecting them directly through the MTA is the best choice.
